BESTSCOPE: Polarizing Microscope
- Polarizing microscope has 2 polarizing filters set at right angles to each other; the first polarizer filter (located before the sample) converts the microscope’s light source into plane-polarized light and an analyzer (second polarizer filter that is positioned after the sample, typically between the objective lens and eyepiece) that is rotated 90° relative to the first polarizer filter (this configuration is also known as “crossed polarizers”).
- When the two polarizing filters are perfectly crossed with no specimen on the stage, the field of view appears dark as the analyzer blocks all the plane-polarized light transmitted by the first filter.
- Isotropic Materials (like glass or liquids) have the same optical properties in all directions and do not alter the polarized light. They appear dark against black background when viewed between crossed polarizers.
- Anisotropic Materials appear bright and sometimes coloured as they alter the light’s polarization. Rotating the circular stage causes transition between bright and dark states on these materials that review different aspects of their internal structure.
